| 背景資料 | Glyceraldehyde 3 phosphate dehydrogenase (GAPDH) is well known as one of the key enzymes involved in glycolysis. As well as functioning as a glycolytic enzyme in cytoplasm, recent evidence suggests that mammalian GAPDH is also involved in a great number of intracellular proceses such as membrane fusion, microtubule bundling, phosphotransferase activity, nuclear RNA export, DNA replication, and DNA repair. During the last decade a lot of data appeared concerning the role of GAPDH in different pathologies including prostate cancer progression, programmed neuronal cell death, age related neuronal diseases, such as Alzheimer's and Huntington's disease. GAPDH is expressed in all cells. It is constitutively expressed in almost all tissues at high levels. There are however some physiological factors such as hypoxia and diabetes that increase GAPDH expression in certain cell types. GAPDH molecule is composed of four 36kDa subunits. |
多克隆抗體
抗原通常是由多個抗原決定簇組成的，由一種抗原決定簇刺激機體，由一個B淋巴細胞接受該抗原所產(chǎn)生的抗體稱之為單克隆抗體（Monclone antibody）。由多種抗原決定簇刺激機體，相應(yīng)地就產(chǎn)生各種各樣的單克隆抗體，這些單克隆抗體混雜在一起就是多克隆抗體（Polyclonal antibody），機體內(nèi)所產(chǎn)生的抗體就是多克隆抗體。
由于多克隆抗體通常包含抗某一抗原的不同表位的抗體，所以，在深固定的樣本中也會很好地發(fā)揮效應(yīng)，在石蠟包埋組織切片的染色中，多克隆抗體選用。根據(jù)實驗的不同需要，多克隆抗體應(yīng)用于相應(yīng)抗原的標(biāo)記。此外，在農(nóng)業(yè)生產(chǎn)中，多克隆抗體被用于農(nóng)藥殘留現(xiàn)場監(jiān)測；在臨床應(yīng)用中，多克隆抗體主要用于病原物的檢測、疾病的診斷及治療，如作為蛋白類免疫抑制劑用于移植反應(yīng)和自身免疫病的治療。
